Genetic Study Links Irish Folklore Giants to Rare Mutation

Genetic research conducted by a collaboration of leading institutions has uncovered a potential explanation for the legends of giants in Irish folklore. The study, which involved Barts and the London School of Medicine and Dentistry, Queen Mary University of London, and other universities including Exeter, Belfast, and Dublin, focused on patients with a hormonal disorder known as acromegaly. Researchers analyzed DNA samples from the general public to identify carriers of a gene linked to childhood-onset acromegaly, which can lead to gigantism.

The research team undertook a comprehensive investigation across Northern Ireland and the Republic of Ireland, involving the active participation of patients and the public. They discovered a notable mutation, referred to as the AIP mutation (R304*), which was found at an unexpectedly high frequency in the Mid-Ulster region. This genetic anomaly appears to be prevalent among the Irish population, as the data suggests that all identified carriers—comprising 18 families and 81 individuals—are descendants of a single ancestor who lived approximately 2,500 years ago.

Among the identified carriers, 31 individuals have developed acromegaly, with over half of these, specifically 18 patients, exhibiting symptoms of gigantism. The researchers emphasized the clinical significance of their findings, stating, “We can now screen family members, and carriers can be followed to pick up the disease early.” Their larger study revealed that 24% of seemingly unaffected gene carriers displayed early signs of acromegaly, leading to immediate surgical interventions for some.

The implications of this research extend beyond medical findings, as the team posits that it may provide a scientific basis for the numerous Gaelic myths surrounding giants in Ireland. The famous Giant’s Causeway and tales of giant creations are among the narratives deeply rooted in Irish culture. Historical figures such as Charles Byrne, whose skeleton was studied in the Hunterian Museum, London, have also been linked to this genetic mutation. Similarly, records indicate numerous giants, including Mary Murphy, known as the ‘Portrush Giantess’, and James Kirkland, one of the ‘Potsdam Giants’, who lived in the region over the centuries.

Professor Sian Ellard from the University of Exeter Medical School, a collaborator on this research, noted, “Irish folklore has numerous stories regarding Irish giants, and the remains of some of these giants have been studied in the past.” She pointed out that this study supports observations made by renowned anthropologist James C. Prichard in 1826, who noted the prevalence of “men of uncommon stature” in Ireland compared to Britain, suggesting an inherent peculiarity in the Irish population.

The researchers also highlighted the possibility that an estimated 436 carriers and 86 affected individuals may remain undiagnosed in Ireland and among people of Irish descent globally. This discovery opens the door to early intervention strategies that could prevent the onset of gigantism and address the associated health risks related to this condition.
